My manager falsely accused me of going into my coworker’s purse to try to steal something. She says this happened months ago but is bringing the complain to me now because my coworker was too shy to say anything. We’ve had a disagreement recently so I believe she may be using this bizarre accusation to try to fire me. How can I defend myself against this? It’s her word against mine.

I would trust her significantly less. if you two were as close as you seem to be under the illusion of, she would bring this up with you in person and not get hr involved and jeopardize your entire career.
If you are innocent maintain your innocence and unequivocally deny any accusation. Ask for evidence of any wrongdoing. If things start going south tell HR and your manager that you will be suing for wrongful termination, trauma, harassment and hostile work environment. Wtf could you even steal from someone’s purse these days?
