The whole point of chappelle's special is that white trans indeed is more powerful than black. Only black guy that can speak on it is dave chappelle or else you're canceled. Even then people will still try to cancel him.
So she was fired for stirring up shit internally and not for the tweet, sounds fair enough. Just because you're offended doesn't mean you can do what you want in a professional setting
Yeeeahh, this person has definitely not watched the specials. Which, sadly, drives home the whole point he's trying to make. The irony of which will be lost on many.
I would argue that, based off the special, Dave doesn't hold the trans community responsible for the suicide of his friend. I believe he admits that he doesn't know what Daphne was feeling/thinking/going through at the time, but posits that the harassment by some vocal members of Twitter certainly didn't help.
I think his anger is pointed towards the toxicity that "Cancel Culture" has become, with everybody on the internet hopping on whatever anger bandwagon they can without contextualizing whatever was said or done. There's been a few comedians pushing back against the knee-jerk reactionary nature of people in the web by giving them the middle finger to show that they won't be intimidated.
I'm sure that Dave is sad that doing so likely hurt a dear friend of his.

Wondering, what would have happened if the employee was a black lesbian… That would have been a tough case. A true clash of Titans.
Trans community: “he hates trans!!”
